Objectives
Contents The teaching module aims to elucidate the main procedures for fixing, processing, and staining of the histological samples, providing the methodological bases regarding the ancillary techniques currently used in the Pathology lab, with particular attention reserved to the immunohistochemical and molecular biology techniques (i.e., in situ hybridization techniques) applied to the clinical and research fields.
Extended Syllabus - Optical Microscopy
- Procedures for acceptance of the biopsy, and macroscopic examination of the surgical samples, with particular attention reserved to the extemporaneous examination
- General principles of the main techniques of fixation, of tissue processing, of the embedding procedures, and sectioning of the paraffin-embedded blocks
- Sectioning and staining protocols
- Staining methods: particular attention will be reserved to the hematoxylin and eosin and some of the frequently used histomorphological, histochemical and special staining techniques
- Tissue processing: pitfalls and technical artifacts
- Principles of the immunohistochemical staining
- How to set up a histological sample for the immunohistochemical staining
- Types of antibodies and methods for the production of monoclonal and polyclonal antibodies
- Physico-chemical properties of the antigen-antibody binding: causes of the antigen unbinding and methods to optimize the use of the immunoreagents
- Antigen identification through immunodetection systems
- Antigen retrieval methods: procedures using heat and protein digestion
- Principal immunohistochemical staining methods
- Multiplexing
- Pitfalls and technical artifacts in immunohistochemistry
- Application of immunohistochemistry in human pathology
- Tissue Microarray (TMA) sample preparation
- Methods for automatic sample preparation and analysis
- Diagnostic roles of the molecular biology techniques (i.e., FISH, CISH, DNAscope, and RNAscope methods) in human pathology
Recommended Bibliography The recommended texts are listed below:
- Bancroft "Theory and Practice of Histological Techniques", 8th Edition, Elsevier (2018)

The teaching slides and the related scientific publications (i.e., “open-access” publications) will be made available for the student after the lessons.
Teaching Methods Lessons provided in the classroom by frontal teaching
Evaluation methods Verification of learning:
The course will be held through frontal lessons. The student preparation will be tested through oral questions, with the final exam score expressed in thirtieths. The student must have learned the general principles of the histochemical and immunohistochemical methods and the main molecular biology techniques applied to the human tissues.
Contacts/More Information The reception of the students will be provided by the teacher upon appointment to be requested via “Teams” platform or via the following e-mail address: rossano.lattanzio@unich.it
